GLP-1 Drugs May Extend Healthspan Through Effects on Epigenetic Aging

A perspective in Experimental Gerontology examines whether GLP-1 receptor agonists—the class of drugs that includes semaglutide (Ozempic/Wegovy) and tirzepatide (Mounjaro)—might extend healthspan by influencing epigenetic aging markers. These drugs are already proven to promote weight loss and improve cardiovascular outcomes in large trials, but this review explores whether their benefits extend to fundamental aging processes. The idea is that GLP-1 drugs might influence DNA methylation patterns and other epigenetic marks associated with biological aging, potentially explaining their broad health benefits beyond glucose control and weight loss. This is still largely speculative—we need actual studies measuring epigenetic clocks in people taking these medications over time. However, the hypothesis is testable and important: if true, it would position GLP-1 agonists as among the first widely-available pharmaceuticals that might genuinely slow biological aging rather than just treating specific diseases. Given millions are already taking these drugs, retrospective analyses of aging biomarkers in existing trial data could provide quick initial answers.
